Adeno-associated Virus (AAV)Due to its good safety, wide range of host cells, and long expression time of exogenous genes in vivo, it is currently one of the viral vectors used in most gene therapies.AAV is widely present in nature and has thousands of serotypes, so AAV antibodies are already present in most humans and mammals.

In order to better predict potential immune responses, optimize and improve the design of preclinical and clinical time to assist in individual screening and matching, and evaluate the pre stored anti AAV antibodies in individuals, it is crucial in gene therapy research. This evaluation improves the safety and effectiveness of clinical testing while ensuring accurate interpretation of clinical trial results, which is beneficial for the development and implementation of gene therapy. In addition, in preclinical studies,Animals must also undergo pre-existing screening for anti AAV antibodies to provide a basis for inter - and intra species selection.

At present, various detection methods for screening anti AAV antibodies have been developed. These methods are generally based on cell transduction inhibition (TI) assays, commonly referred to as neutralizing antibody (NAb) assays, to evaluate the effects of neutralizing antibodies (NAb). In addition, there is total antibody (TAb) assay, also known as binding antibody assay, which identifies all AAV antibodies that bind to the capsid, regardless of their neutralizing ability. The determination of TAb is widely used for large-scale sample screening of anti AAV antibodies due to its fast speed, high sensitivity, and easier operation.
The detection methods for TAb include antigen capture method and bridging method, and the detection forms of the two methods are as follows:

01 Long development time for analytical methodsSpecific analysis methods need to be developed for different serotypes and species of AAV, and reliable methods need to be provided to regulatory authorities, which is time-consuming and labor-intensive.
02 AAV needs to be labeledThe current TAb detection method requires the use of AAV capsids of specific serotypes for coating and labeling, which requires a large amount of manual operation, increases the risk of detection variability, and reduces the reproducibility of data; Additionally, it requires the consumption of a large amount of expensive AAV capsids, resulting in high detection costs.
03 Large scale sample screening requires a significant investment of manpowerLarge animals (especially NHPs) are widely exposed to AAV and may have pre-existing antibodies that cross react with AAV serotypes. To ensure the effectiveness of the model and correct interpretation of preclinical results, it is necessary to screen a large number of samples for anti AAV antibodies.
04 The traditional ELISA method consumes a large amount of samples, resulting in a decrease in the yield of the final product.

The Gyrolab universal anti AAV antibody detection kit simplifies the detection process of AAV pre stored antibodies,Suitable for both Cyno and human sample detection.
