Extending the lifespan of reagents: key points for environmental control and reagent management of primer synthesizers

The reagent lifespan of primer synthesizers directly affects synthesis efficiency and cost, while environmental temperature and humidity, light exposure, oxidation, and microbial contamination are the main factors leading to reagent degradation. Through refined environmental control and reagent management, the validity period of reagents can be significantly extended and the synthesis failure rate can be reduced.
1、 Environmental control: blocking external degradation factors
temperature management
The reagents should be stored in a low temperature environment of 2-8 ℃ to avoid high temperature accelerating the hydrolysis of phosphoramidite monomers. The internal reaction module of the synthesizer should be equipped with an independent temperature control system to ensure stable coupling reaction at 20-25 ℃ and prevent temperature fluctuations from causing a decrease in reagent activity. For example, a certain model of synthesizer controls the temperature fluctuation of the reaction chamber within ± 0.5 ℃ through semiconductor refrigeration chips, reducing reagent consumption by 15%.
Humidity and lighting control
Excessive humidity can cause the reagent to absorb moisture and form lumps, while light exposure (especially ultraviolet light) can damage the photosensitive protective groups. It is recommended to use a sealed reagent chamber with built-in desiccant and shading coating, combined with a nitrogen protection system, to maintain the humidity inside the chamber at<30% RH and oxygen concentration<1%. A laboratory has extended the storage time of phosphoramide monomer from 3 months to 9 months through this scheme.
Microbial pollution prevention and control
If microorganisms are mixed into the reagent, their metabolites will catalyze the decomposition of the reagent. Regular high-temperature and high-pressure sterilization (121 ℃, 30 minutes) of the reagent pipeline is required, and disposable sterile reagent bottles should be used. For open synthesizers, a 0.22 μ m filter membrane can be installed to intercept particulate matter and microorganisms in the air.
2、 Reagent management: optimizing the usage process
Packaging and dark storage
Large capacity reagent bottles are prone to contamination due to repeated use after opening. It is recommended to divide them into brown opaque bottles according to the single synthesis amount, and seal the remaining reagents before refrigeration. For example, dividing 50mL of phosphoramide monomer into 5mL/bottle can reduce the exposure risk by 90%.
Real time monitoring and early warning
By monitoring the pH, conductivity, and color changes of the reagent through sensors, an automatic alarm will be triggered when the data deviates from the threshold. The intelligent reagent management system developed by a certain enterprise can predict the risk of reagent failure 3 days in advance, avoiding synthesis failure caused by reagent deterioration.
Inert gas protection
Continuously introduce high-purity nitrogen gas (purity>99.999%) into the reagent delivery pipeline to isolate oxygen and moisture. Tests have shown that nitrogen protection can extend the activity retention time of coupling reagents from 72 hours to 168 hours.
3、 Effect verification and cost-benefit analysis
After a third-party testing institution implemented the above plan, the reagent waste rate decreased from 25% to 8%, and the cost of synthesizing a single primer decreased by 40%. Meanwhile, the synthesis failure rate caused by reagent degradation decreased from 12% to 2%, significantly improving experimental efficiency.
Through the collaborative optimization of environmental control and reagent management, the reagent life of the primer synthesizer can be extended by 3-5 times, providing stable support for high-throughput synthesis, clinical diagnosis and other scenarios. It is recommended that users regularly calibrate environmental monitoring equipment and establish reagent usage records to continuously optimize management strategies.
