The water quality analysis photometer is a core device based on Lambert Beer's law, widely used for quantitative detection of specific substances (such as heavy metals, nutrients, organic matter, etc.) in water quality. The measurement accuracy is affected by multiple factors, and the following analysis is conducted from four aspects: instrument characteristics, sample properties, experimental conditions, and operating standards:
1、 Performance limitations of the instrument itself
1. Light source stability
Light sources such as tungsten lamps/deuterium lamps decay over time, causing fluctuations in incident light intensity and directly affecting the repeatability of absorbance;
Using a regulated power supply can alleviate the light intensity drift caused by voltage fluctuations.
2. Resolution of monochromator
Excessive spectral bandwidth of filter or prism systems can introduce stray light, reducing measurement specificity at the target wavelength;
The high-precision instrument is equipped with a narrowband interference filter (full width at half maximum<5nm), which significantly improves the anti-interference ability.
3. Sensitivity of detector
The dark current noise and fatigue effect of photomultiplier tubes (PMT) can cause signal distortion in low concentration samples;
Regularly calibrating the detector gain can maintain a linear response range.
2、 Sample matrix complexity
1. Turbidity and suspended solids
The scattering of incident light by particulate matter generates a pseudo absorption signal, which needs to be eliminated by pre-processing (0.45 μ m filter membrane) to eliminate interference;
It is recommended to add coagulants to the colloidal system for precipitation and then centrifuge for separation.
2. Color background interference
Humus and other substances in natural water bodies come with yellow/brown colors and form background absorption in the visible light range;
The dual wavelength difference subtraction method can effectively deduct such background interference.
3. Coexistent ion effect
High concentration salts can alter the dielectric constant of the solution and affect the color rendering efficiency of the complex;
Matrix matching method (blank control) can correct differences in ion strength.
3、 Kinetic control of colorimetric reaction
1. pH sensitivity
Most color reagents (such as ortho phenanthrene for iron measurement) are sensitive to pH and require strict control of the buffer system;
A pH deviation of ± 0.2 from the optimal value may lead to the decomposition of the complex or the occurrence of side reactions.
2. Color rendering time window
The generation of colored substances requires a certain reaction time, and reading too early/too late can cause errors;
Draw an absorbance time curve to determine the stable plateau period (usually 10-30 minutes).
3. Temperature dependence
The color reaction rate increases with temperature, but high temperature may accelerate fading;
Constant temperature water bath control within ± 1 ℃ can ensure reaction consistency.
4、 Operation and environment variables
1. Differences in colorimetric dishes
The optical path accuracy (± 0.1mm) and scratches on the transparent surface of glass/quartz materials introduce systematic errors;
Use matching colorimetric dishes and regularly verify the optical path length.
2. Environmental light interference
Natural light in the laboratory contains UV visible components, and direct exposure to the sample cell can cause readings to be higher;
The use of a light shield in conjunction with darkroom operation can eliminate the influence of stray light.
3. Personnel operation variation
Volume errors in pipetting (such as pipette calibration deviation>1%) are directly amplified to the final result;
Standardized SOP processes can reduce human operational differences.
