High throughput tissue grinderThe operation process mainly includes the following steps:
Instrument preparation:
Place the instrument on a dry, ventilated, and stable experimental platform, avoiding enclosed or humid environments.
Ensure good grounding of the instrument, connect to a 220V power supply, and check if the control display lights up normally.
Test the function of the control button and confirm that there is no abnormal noise during no-load operation.
Check if the fixtures, adapters, grinding tanks, and other components are securely fastened, and if the springs and housing bolts are not loose or damaged.
Sample preparation:
Select the appropriate size of grinding jar or centrifuge tube adapter based on the sample type (such as plant tissue, animal tissue, fungi/bacteria, etc.).
The sample size should be controlled within 1/3 of the container volume, and an appropriate amount of grinding beads (such as stainless steel beads or ceramic beads) should be added.
If low-temperature grinding is required, freeze the adapter containing the sample in liquid nitrogen for 1-2 minutes (or more than 3 minutes), and be careful not to directly contact the alloy fixture with liquid nitrogen.
Loading samples:
Place the frozen adapter or grinding tank into the fixture and secure it with a vertical positioning knob and a horizontal positioning slot to ensure the safety lock is in place.
For centrifuge tube adapters, the sample tubes should be symmetrically placed to maintain the balance of the grinding table and avoid unbalanced operation.
Parameter settings:
Set the running time and frequency of the grinder. For example, the recommended frequency for grinding plant leaves is 1000-1100r/min, with a duration of 1 minute; Animal tissues (such as liver) are recommended to have a frequency of 1000-1600r/min and a duration of 1-3 minutes.
For difficult to break samples (such as seeds), a higher frequency (such as 1200-1600r/min) can be set and the grinding can be repeated 2-3 times.
Start grinding:
Close the safety door and confirm that the parameters are correct.
Press the "START" button, and the instrument will start running. The real-time frequency and remaining time will be displayed on the control panel.
If an emergency stop is required, press the "STOP" button (but frequent operation is not recommended).
Grinding completion and sample collection:
After the instrument stops automatically, open the safety door, turn the vertical fixing knob counterclockwise, and remove the adapter or grinding tank.
If the grinding is not good, you can repeat the steps of fixing, parameter setting, and starting.
Instrument cleaning and maintenance:
Turn off the power, clean the grinding table and adapter to prevent sample residue from corroding the instrument.
Regularly inspect the wear of components such as grinding beads and fixtures, and maintain them according to laboratory standards.
Safety precautions:
Wear protective equipment (such as gloves and goggles) during operation, especially during the liquid nitrogen pre cooling step.
It is strictly prohibited to operate the instrument in an unbalanced state to avoid damaging the equipment or affecting the grinding effect.
