solid-phase extractionSPE technology, as the core means of sample pretreatment, achieves target enrichment and matrix purification through selective adsorption and elution. Its performance directly determines the sensitivity and accuracy of subsequent analysis. This article analyzes the core technical system of solid-phase extraction instrument from three dimensions: adsorbent materials, flow rate control, and automated process design.
1、 Adsorbent material: the cornerstone of selective adsorption
Adsorbents are the core of solid-phase extraction, and their types and properties determine the retention efficiency and anti-interference ability of the target substance. According to their mechanism of action, adsorbents can be divided into the following four categories:
Silicone based adsorbent
Representative types: C18, C8, NH2, CN and other bonded silica gel.
Technical principle: By bonding with organic functional groups such as octadecyl and amino groups through silicon hydroxyl groups, non-polar or polar stationary phases are formed. For example, C18 fillers are suitable for the extraction of non-polar compounds such as polycyclic aromatic hydrocarbons due to their long carbon chains and strong hydrophobicity; NH2 fillers have both positive and negative ion exchange interactions and are commonly used for the separation of sugars and organic acids.
Optimization direction: To address the interference of residual silicon hydroxyl groups on the surface of silicone gel, trimethylchlorosilane is used for tail sealing treatment, which can reduce non-specific adsorption of polar impurities and improve recovery rate.
Inorganic matrix adsorbent
Representative types: alumina, Florisil, graphitized carbon black.
Technical principle: Utilize surface active hydroxyl groups or special structures to achieve adsorption. For example, alumina adsorbs polar compounds through hydrogen bonding or ion exchange, and its activity can be controlled by pH adjustment (acidic alumina pH ≈ 5, alkaline alumina pH ≈ 8.5); Graphitic carbon black is suitable for extraction of polar and non-polar mixtures due to its positive surface charge, hydrophobicity, and anion exchange ability.
Application scenario: Florisil is commonly used to extract polar pesticide residues from non-polar substrates such as vegetable oils.
Organic polymer adsorbent
Representative types: HLB (hydrophilic lipophilic balance polymer), MCX (strong cation exchange polymer).
Technical principle: By providing a large specific surface area through a three-dimensional network structure, broad-spectrum adsorption can be achieved. For example, HLB fillers have both hydrophilic and hydrophobic groups, making them compatible with solvents across the entire pH range and suitable for extracting complex matrices such as drug metabolites and environmental hormones; MCX fillers achieve selective retention of alkaline compounds through sulfonic acid groups.
Technical advantages: Compared to silicone matrix, polymer adsorbents have higher chemical stability, are resistant to acid, alkali, and organic solvents, and have an extended service life of over 500 times.
Specialized adsorbent
Representative types: azo dye specific column, aminoethyl ester specific column.
Technical principle: Design functional groups for specific target substances. For example, azo dye specific columns selectively adsorb dye molecules containing aromatic rings through π - π interactions, meeting the requirements of the GB/T 17592-2011 standard.
Industry value: Specialized columns can simplify the method development process and reduce the detection limit to the 0.1ppb level.
2、 Flow rate control technology: precise regulation of adsorption and elution processes
The stability of flow rate directly affects the recovery rate and repeatability of the target substance. Traditional manual SPE relies on manual adjustment, resulting in flow rate fluctuations of up to ± 2mL/min and a recovery rate deviation of ± 15%. Modern solid-phase extraction instruments achieve precise flow rate control through the following technologies:
High precision CNC pump
Technical principle: Real time adjustment of pump speed using PID algorithm, combined with high-precision metering pump (flow accuracy ≤ 1%), ensuring flow rate fluctuation<± 0.5mL/min.
Technical advantage: The linear correlation coefficient between flow rate and rotational speed is ≥ 0.999, which avoids the penetration or overload of the target object caused by too fast flow rate.
Positive pressure injection technology
Technical principle: The sample is driven through the extraction column by gas pressure (stable ± 0.1MPa), replacing the traditional negative pressure suction method.
Technical advantages: The positive pressure system can eliminate dead volume in the flow path, reduce sample residue, and is particularly suitable for the extraction of thermosensitive samples (such as plasma drugs).
Valve switching technology
Technical principle: Using an electromagnetic valve group instead of a robotic arm to achieve rapid flow path switching (response time<0.5 seconds).
Technical advantages: Avoiding mechanical failures and supporting multi-channel parallel processing (such as simultaneous extraction of 6 channels), reducing the processing time for a single batch from 4-8 hours to 90 minutes.
3、 Automated Process Design: From Manual Operations to Intelligent Standardization
Automation is a solid-phase extraction instrumentThe core development trend is that its design needs to cover the entire process of sample activation, loading, rinsing, and elution, and integrate intelligent control and safety protection functions.
Full process automation control
Technical implementation: Pre stored 20+standard methods (such as pesticide residue and veterinary drug residue detection) through a graphical user interface, supporting automatic/step-by-step operation modes. For example, users only need to input the quantity and type of samples, and the instrument can automatically complete the following steps:
Activation: Rinse the extraction column with methanol and water in sequence to remove impurities and activate adsorption sites;
Sample loading: The sample solution is passed through the extraction column at a controllable flow rate (such as 10mL/min), and the target substance is selectively retained;
Rinsing: Use strong polar solvents (such as 5% methanol-95% water) to remove interfering components;
Elution: Use strong solvents such as acetonitrile to elute the target substance and collect it.
Technical advantages: Single batch processing time is reduced to 1/4 of traditional methods, and the recovery rate remains stable at 92% -105% (RSD<3%).
Intelligent security protection system
Overpressure protection: Automatic shutdown alarm when pressure>0.3MPa to prevent extraction column rupture;
Overtemperature protection: When the temperature exceeds 50 ℃, the cooling system is triggered to ensure the stability of thermosensitive samples;
Automatic cleaning program: supports custom cleaning process of "5mL water rinse → 5mL methanol rinse", combined with automatic rinsing function between samples, reducing cross contamination rate to<2%.
Modular and Scalable Design
Technical implementation: The equipment comes standard with 6 fixed brackets (hole spacing 5cm), compatible with mainstream extraction columns of 1mL, 3mL, 6mL, and 12mL; Simultaneously supporting the integration of online concentration modules (such as nitrogen blowing concentration), achieving the integration of "extraction concentration analysis".
Application scenario: A certain environmental protection laboratory uses a solid-phase extraction instrument with an integrated online concentration module to compress the detection cycle of polycyclic aromatic hydrocarbons in 10L water samples from 5 days to 2 days.
