What details should be paid attention to when using packed chromatography columns
Date: 2025-10-22Read: 1
The packed chromatographic column is the core component of chromatographic analysis, and its performance directly affects the separation efficiency and detection accuracy. The scientific and standardized usage process can significantly improve column efficiency and extend service life. The key points are explained in detail from four aspects: pre loading preparation, sample loading operation, operation management, and maintenance.
1、 Pre treatment before loading
Column screening and cleaning
Select suitable fillers (such as non-polar C18 and polar silica gel) based on the polarity of the target substance. The new and old chromatography columns need to be cleaned with methanol/acetonitrile ultrasound for 30 minutes to remove any residual preservatives.
Soak the empty column tube in dilute hydrochloric acid for 2 hours, rinse with pure water until neutral, and then rinse with methanol to ensure that there are no hydrophobic impurities interfering with the inner wall.
Preparation and loading of homogenate
When using the settling method for filling, the filler is made into a uniform suspension (with a solid content of about 5%), and the filler is tightly packed by suction with a vacuum pump; Dry filling requires layered tapping of the cylinder to eliminate gaps.
Install sieve plates at both ends of the column bed (aperture<1/3 of the packing particle size) to prevent packing leakage, and fix the top with glass wool.
2、 Sample loading and operation control
Balance and activation
When using a new column, rinse with a low initial flow rate of 0.5mL/min for 2 hours to remove any bubbles; The reverse phase column needs to gradually increase the proportion of organic phase to the working conditions to avoid the collapse of the stationary phase.
Calibrate with blank mobile phase baseline before injection, and start the experiment after the UV detector signal stabilizes.
Principle of parameter optimization
Flow rate control: Maintain the optimal linear velocity (usually 1-2mm/s for HPLC), too high will intensify vortex diffusion, and too low will increase mass transfer resistance.
PH range limitation: Silicone matrix should avoid pH>8, and polymer fillers have a wider tolerance range, but special pH accelerates the breaking of the sealing bond.
Temperature management: Raising the column temperature can improve peak shape, but exceeding 60 ℃ can easily cause bond phase loss, and precise temperature control with a temperature control module is required.
3、 Daily maintenance and regeneration
Pollution response strategies
Gradient elution of lipid soluble pollutants (such as methanol → dichloromethane → methanol); Ionic residues were washed with a salt solution (0.1M NaCl).
When there is severe blockage, remove the column head, remove the upper layer of deteriorated filling material, replenish fresh filling material, and re compact.
Long term storage standards
Short term idle (<1 week): sealed in a closed container containing mobile phase, with electricity flushing for 30 minutes per week;
Long term storage (>1 month): Replace with a mixture of acetonitrile/isopropanol (1:1), seal both ends, and place upright in a cool place.
4、 Key precautions
Prohibition of sudden changes: When switching the mobile phase, a stepped transition is required to prevent sudden changes in solvent viscosity from causing severe fluctuations in column pressure; Replace the buffer salt solution with an inert solvent (such as acetonitrile) before stopping the machine.
Sample pretreatment: Complex biological samples need to be purified by SPE columns to avoid protein precipitation blocking the column bed; Injection volume ≤ 1% of column capacity, overloading leads to severe tailing.
Abnormal phenomenon diagnosis: Continuous increase in column pressure indicates particle accumulation, baseline drift suggests two-phase imbalance, and ghost peaks are mostly caused by system pollution.
Through refined operation and regular maintenance, the theoretical number of trays for filling the chromatographic column can be maintained at over 90% of the initial value, with a single analysis time deviation of less than 2%. In practical applications, it is necessary to establish a standardized process based on specific packing characteristics and record the column efficiency attenuation curve for timely replacement.
