The following is a systematic analysis and solution for the problem of "inverted peak" (i.e., the baseline protrudes upwards to form a reverse peak shape) in gas chromatographs, covering cause diagnosis, troubleshooting steps, and preventive measures to help quickly locate and solve the problem:
1、 The core cause of peak inversion
The essence of peak inversion is that the detector response value is opposite to the expected direction, mainly caused by the following two factors:
Abnormal physical mechanism: instantaneous signal reversal caused by compressibility differences or thermodynamic effects in the sample or carrier gas;
Operational/equipment defects: injection errors, detector contamination, circuit signal interference, or incorrect data processing parameters.
2、 Step by step investigation and solution
1. Preliminary observation and recording
-Reproduction verification: Repeat the injection of blank solvent and standard solution to confirm whether the peak inversion continues to occur. If only a specific sample appears, it indicates that the sample has special properties; If it exists universally, focus on the instrument itself.
-Compare normal spectra: Retrieve historical normal spectra, compare retention time, peak area, and baseline morphology to determine whether it is a single peak anomaly or global distortion.
2. Sample injection system troubleshooting
-Contamination of spacer debris: The detachment of old injection spacer debris may clog the liner, causing a sudden change in pressure during injection.
Replace the new spacer, cut a section of the waste lining pipe at the front end, and reinstall it.
-Excessive injection volume: Overloaded injection causes incomplete volatilization of the sample in the vaporization chamber, leading to nonlinear response caused by cold focusing effect.
Reduce the injection volume (e.g. from 1 μ L to 0.5 μ L), or dilute the sample before injection.
-Misalignment of diversion ratio: blockage of diversion outlet or malfunction of regulating valve, resulting in actual diversion ratio lower than the set value.
Check if the diversion outlet pipeline is bent/blocked, and clean the diversion plate with solvent ultrasonic cleaning.
3. Carrier gas and flow path inspection
-Insufficient purity of carrier gas: Impurities such as oxygen and moisture react with the sample, altering the detector's response characteristics.
Replace the high-purity carrier gas cylinder and install a gas purifier (such as a deoxygenation tube).
-Pressure fluctuation in front of the column: EPC (Electronic Pressure Control) module failure or gas path leakage, resulting in sudden changes in flow rate.
Perform a column front pressure test, observe the smoothness of the pressure curve, and tighten the nuts at each interface.
-Column efficiency decline: The aging chromatographic column loses the stationary phase, resulting in a decrease in separation efficiency accompanied by ghost peaks.
Cut off the front end of the column by 10-30cm and re age it. If it is severe, replace the column with a new one.
4. Special diagnosis for detectors
-Typical issues with FID detectors:
-Imbalance of hydrogen/air ratio: Improper gas ratio leads to unstable flames and generates electrical noise.
Reset H ₂: Air=1:10 according to the instructions. After igniting the flame, observe that the color should be light blue.
-Collecting carbon deposits: Long term high concentration sample deposition leads to signal drift.
Disassemble the collection electrode, clean it with isopropanol ultrasonic cleaning, and then dry it.
-Attention points for TCD detector: The temperature difference between the reference arm and the measuring arm is too large, or the hot wire current is set too high, which may cause thermal noise.
Calibrate the temperature difference of the tank body to be less than 0.1 ℃, and reduce the bridge flow to the recommended lower limit.
5. Optimization of data collection parameters
-Signal inversion setting: Some workstations provide the option to display "peak inversion", which can cause visual inversion if accidentally turned on.
Check the signal polarity settings of the data processing software and restore the default forward mode.
-Excessive filtering parameters: Excessive digital filtering can distort the true peak shape.
Reduce the cut-off frequency of the high pass filter appropriately (such as from 0.1Hz to 0.01Hz).
Through the above systematic investigation, the source of the peak inversion can be quickly located. If the problem still cannot be solved, it is recommended to contact an engineer for in-depth diagnosis, focusing on checking the internal electrode connections of the detector and the signal grounding of the circuit board. Standardized daily operations and regular maintenance are key to avoiding such problems.
