Methanol, acetonitrile, and phosphate buffer are commonly used mobile phases. We used a reverse phase C18 chromatographic column.
1. After analyzing the sample and cleaning the chromatographic column, connect a PEEK tube to the original position where the column was connected. Clean with deaerated pure water at a flow rate of 1.5mL/min for 1.5 hours, and then clean with methanol under the same conditions. Replace the cleaning solution of the automatic sampler with methanol, open the drain valve, use a syringe to aspirate more than 50mL, and then close the drain valve, purge and rinse three times each. After completion, press shift and 9 to close the automatic sampler.
Pure water cleaning chromatography system can remove residual buffer salts in the pipeline, methanol cleaning can prevent the system from breeding bacteria and microorganisms blocking the pipeline, and can also wash away insoluble substances in water.
Precautions during the use of an automatic sampler:
1) Before use, check whether there are cracks in the syringe of the sampler and whether the needle tip is hairy;
2) Remove residual samples from the injector, wash the injector with professional solvents about 5-20 times, and discard the waste liquid from the first 2-3 times;
3) Immerse the needle in the solvent, remove any bubbles from the injector, and repeatedly extract the sample. When excluding samples, the bubbles in the injector can change with the vertical variation of the tube;
4) While using the injector, fill the injector with liquid and then remove the liquid to the desired volume for injection.
Regarding the cleaning process of the automatic sampler;
The cleaning reagents commonly used in laboratories are selected based on the pollutants, and dichloromethane, methanol, acetonitrile, and acetone are commonly used. When cleaning the injector, do not block the needle. Remove the plunger, inject the cleaning solvent into another injector, and then insert the plunger to gently push the solvent out of the needle.
Automatic sampler disinfection mode: Use high-pressure sterilization method to disinfect the sampler while extracting the plunger, and use ethylene oxide;
During the process of soaking the injector in solvent, the entire injector should not be soaked in the solvent, as this can easily damage the adhesion of the bonding parts on the injector. Use cotton paper or thin paper when cleaning the outside of the sampler.
