Carbon dioxide incubator is a key equipment for cell culture, widely used in biomedical research, pharmaceuticals, clinical diagnosis and other fields. The following are typical use cases and operational points:
1、 Common application scenarios
1. Cell culture
Case: Cultivate HeLa cells for cancer research.
Parameter settings: 37 ℃, 5% CO ₂ (simulated in vivo environment), humidity>90% to prevent evaporation of the culture medium.
Key point: Regularly calibrate the CO ₂ concentration sensor to avoid pH fluctuations affecting cell growth.
2. Stem cell research
Case: Inducing differentiation of pluripotent stem cells (iPSCs) into cardiomyocytes.
Special requirements: Strict sterility is required (with built-in UV sterilization or HEPA filters), and O ₂ concentration control is required (some experiments require low oxygen conditions).
3. Virus cultivation and vaccine development
Case: Cultivate Vero cells to produce rabies vaccine.
Attention: A biosafety level incubator (such as a secondary biosafety cabinet) is required to avoid aerosol contamination.
4. Organizational Engineering
Case: 3D printed cartilage tissue matures in an incubator.
Parameter adjustment: CO ₂ can be adjusted to 7 10% to promote collagen synthesis.

2、 Operation precautions
1. Sterilization and Cleaning
Wipe the inner wall with 70% ethanol before use, and regularly sterilize at high temperature (such as 180 ℃ dry heat).
The water tray needs sterile water to avoid microbial contamination.
2. Gas control
CO ₂ steel cylinders need to be equipped with pressure reducing valves, and it is recommended to use gas with a purity of ≥ 99.9%.
After starting up, it is necessary to stabilize for 4 to 6 hours before inserting the cells (to ensure temperature, humidity, and CO ₂ balance).
3. Monitoring and Calibration
Daily recording of temperature and CO ₂ concentration (such as infrared sensor data).
Calibrate the sensor once every quarter (using standard gas).
4. Fault response
Large fluctuations in CO ₂: Check for cylinder pressure, pipeline leaks, or sensor malfunctions.
Insufficient humidity: Check the water level in the water tray or the humidification system.

3、 Special case: Primary cell culture
Challenge: Primary cells (such as liver cells) are sensitive to the environment.
Optimization plan:
Use a pre balanced incubator (set parameters 24 hours in advance).
Add antibiotics (such as penicillin and streptomycin) to prevent contamination.

By setting reasonable parameters and standardizing operations, the carbon dioxide incubator can significantly improve the repeatability and reliability of cell experiments. In practical applications, it is necessary to select functions based on experimental purposes (such as shaker integration, real-time monitoring, etc.).
