Principle and function of fully automatic fat analyzer and fat extractor
The fully automatic fat analyzer uses the Soxhlet extraction principle and weight measurement method to determine fat content. Dissolve fat in organic flux, separate the fat from the flux using extraction method, then dry, weigh, and calculate the fat content. It mainly consists of three parts: heating extraction, flux recovery, and cooling. During operation, the heating temperature can be adjusted according to the boiling point of the reagent and the ambient temperature. The sample is repeatedly soaked and extracted during the extraction process to achieve rapid measurement.
Features:
1. Compact size, fast heating and uniform heating in water bath.
2. All instruments use glass grinding joints to avoid leakage problems.
3. Digital temperature control, temperature control, simple and convenient operation.
Instructions for Use:
1. Before dissolution, the sample should be ground first. The degree of grinding can directly affect the extraction speed of fat. Adequate grinding can better dissolve fat in organic solvents such as petroleum ether.
2. After the sample is ground, place the sample into a filter paper cylinder. At this time, the filter paper cylinder should be tightly packed, and the height of the filter paper cylinder should not exceed the reflux bend, otherwise the solvent is not easy to penetrate the sample.
When extracting, yimi is used as the solvent for the fat, with a boiling point of around 40 ℃, so the temperature should not be too high, usually around 70-85 ℃.
4. The reagent used in the experiment is anhydrous. For example, if yimi is used in the experiment, it should be kept anhydrous because if it contains other substances such as water, it may extract sugars or inorganic substances from the sample, affecting the determination of fat. Therefore, it is also necessary to ensure that the solvent cups and other items used in the experiment are dry.
5. Connect a calcium chloride drying tube to the upper end of the condenser tube, which not only prevents moisture from entering the air, but also prevents Yimi from evaporating into the air, thus preventing air pollution in the laboratory's small environment. If this device is not available, a dry cotton ball can also be stuffed in. Generally, fully automatic or semi-automatic machines will have a lid at the top of the condenser tube, which can also effectively protect the experiment and prevent solvent evaporation.
After fully extracting the fat with a fat analyzer, the sample and ether extract can be dried or the solvent cup can be dried. The drying time should not be too long, as some unsaturated fatty acids are easily oxidized during the heating process, which affects the final result of fat determination. Without a vacuum drying oven, it can be dried at 100-105 ℃ for 1.5-3 hours.
