The working principle of anion concentration column and its application in sample pretreatment
Date: 2025-08-18Read: 1
The core working principle of anion concentration column is based on ion exchange and dynamic adsorption mechanism. The fixed phase is usually a quaternary ammonium functional resin with a positive charge. When a sample solution containing target anions (such as Cl ⁻, NO ⁻, SO ₄ ² ⁻) flows through the column, the cations on the resin surface (such as Na ⁺) are replaced by the anions in the sample. The target anions are adsorbed onto the resin surface through electrostatic interactions, while non target substances (such as neutral molecules or cations) flow out directly with the mobile phase. This process achieved the initial separation of the target anion from the matrix.
The enrichment effect is a key function of the concentration column. By controlling the composition of the mobile phase (such as low concentration eluent) and flow rate, the retention time of the target anion in the column can be extended, allowing it to form a dynamic adsorption equilibrium on the resin surface. For example, when analyzing fluoride ions in surface water, diluting the sample 10 times and passing it through a column can increase the fluoride ion concentration from trace level to above the chromatographic detection limit, meeting the requirements of quantitative analysis. In the elution stage, the salt concentration of the eluent is increased by gradient or the pH is adjusted to disrupt the adsorption equilibrium, allowing the target anions to desorb sequentially according to their affinity differences, achieving efficient separation.
In sample pretreatment, anion concentration columns are widely used in fields such as environmental monitoring, water quality analysis, food testing, and industrial process control. For example, when monitoring nitrate pollution in groundwater, a concentration column can enrich trace amounts of NO ∝⁻ from large volume water samples, eliminate matrix interference, and improve detection sensitivity; In the detection of food additives, pre-treatment of the concentration column can reduce the damage of high salt matrix to the chromatographic column and prolong the service life of the instrument. In addition, its compatibility with automatic samplers supports online continuous analysis, significantly improving laboratory work efficiency.
In practical applications, attention should be paid to the operating procedures: the sample needs to be filtered through a 0.45 μ m membrane to remove particulate matter; High concentration samples need to be diluted to a linear range to prevent resin overload; After elution, rinse with regeneration solution (such as 10% NaOH) to remove residual organic matter. Through standardized operation, the anion concentration column can stably achieve 200-500 injections, providing reliable support for trace anion analysis.
