Real time fluorescence quantitative PCR instrumentIt is an instrument used for fluorescence measurement. This device consists of a fluorescence quantification system and a computer, used to monitor the fluorescence of the cyclic process. Computers connected to real-time devices collect fluorescence data. The data is displayed in the form of charts through real-time analysis software developed. The raw data is plotted as a graph of fluorescence intensity relative to the number of cycles. After collecting the raw data, analysis can begin. The software of real-time devices can normalize the collected data to compensate for differences in background fluorescence. After normalization, the threshold level can be set, which is the level for analyzing fluorescence data.

Plate loading specifications:
Ensure that the hole positions of the reaction plate match exactly with the software template (if the offset is greater than 0.5mm, it needs to be reloaded).
Use optical transparent sealing film (thickness ≤ 0.1mm) to avoid fluorescence signal attenuation.
Key points of program settings:
It is recommended to maintain 95 ℃ for 10 minutes during the pre denaturation stage (to ensure complete denaturation of the template).
Melting curve analysis requires setting up a continuous heating mode (0.5 ℃/s).
Maintenance and Quality Control:
Daily cleaning:
Wipe the sample chamber with 70% ethanol every week (avoid using chlorine containing disinfectants).
Replace the cooling fan filter every 3 months.
Pollution prevention and control:
Experimental partitioning operation (separate pre-processing zone, amplification zone, and analysis zone).
Use an anti contamination system containing uracil DNA glycosylation enzyme (UDG).
