Regeneration of chromatographic column

Liquid chromatography columns are consumables that may experience a decrease in peak height, an increase in peak width, or the appearance of shoulder peaks with increasing usage time or injection frequency, resulting in a decrease in column efficiency. Regular cleaning and regeneration are required.
1. Reverse column
Wash the mobile phase with solvents such as methanol: water=90:10, pure methanol, dichloromethane, etc. in sequence, and each mobile phase should flow through the chromatographic column at least 20 times the volume of the column. Then rinse in reverse order.
2、 正相柱
Use solvents such as n-hexane, isopropanol, dichloromethane, methanol, etc. as mobile phases, wash them sequentially, and each mobile phase flows through the chromatographic column at least 20 times the column volume (isopropanol has a high viscosity, so the flow rate can be reduced to avoid excessive pressure). Be careful not to reverse the order of solvent use. After rinsing with methanol, rinse with n-hexane in the opposite order. All mobile phases must be strictly dehydrated.
3. Ion exchange column
Long term use and injection in buffer solution will lead to a decrease in the ion exchange capacity of the chromatographic column,. Rinsing with dilute acid buffer solution can regenerate the cation column, while rinsing with dilute alkali buffer solution can regenerate the anion column.
Users can also choose solvents that can dissolve pollutants inside the column as the mobile phase for forward and reverse flushing. But the column efficiency of the regenerated chromatographic column cannot be restored to the level of the new column.
If the column is installed upside down, it can be adjusted back, but it may cause the support body inside the column to collapse. Try not to reverse load the chromatography column unless absolutely necessary.
