Introduction to the working principle of liquid chromatography
Date: 2025-08-29Read: 1

Liquid ChromatographIt is a method of using a mixture in liquid-An instrument for separating, analyzing, and identifying mixtures based on the difference in distribution ratio between solid or immiscible liquids.

Liquid ChromatographThe working principle of separation is based on the difference in interaction forces between the fixed phase (filler) and the mobile phase (solvent) of the sample solution. The specific process is as follows:

1.Mobile phase transport: The mobile phase in the reservoir is pumped into the system by a high-pressure pump.

2.Sample injection: The sample solution enters the mobile phase through the injector and is loaded into the chromatographic column (stationary phase) by the mobile phase.

3.Separation process: When the components in the sample undergo relative motion between the stationary phase and the mobile phase, they undergo repeated adsorption-The distribution process of desorption results in significant differences in the movement speed of each component, leading to their separation into individual components that flow out of the column in sequence.

4.Detection and recording: When passing through the detector, the sample concentration is converted into an electrical signal and transmitted to the recorder. The data is printed in the form of a graph for researchers to analyze.

Liquid chromatography is widely used in the following fields due to its efficient separation, high sensitivity, and rapid analysis characteristics:

Pharmaceutical analysis: used for qualitative and quantitative detection, impurity analysis, and quality control of drugs.

Biochemical analysis: plays an important role in proteomics, metabolomics, and other fields, used for the separation and analysis of biomolecules.

Food analysis: detecting additives, pesticide residues, and nutritional components in food.

Environmental analysis: Analyze pollutants such as heavy metals and organic pollutants in environmental samples such as water and soil.
