As a commonly used equipment in laboratories for crushing cells and dispersing tissues, the stability of biological sample homogenizers directly affects the accuracy of experimental results. Scientific maintenance can extend equipment lifespan and reduce failure rates. The following provides a detailed explanation of maintenance details from five aspects: daily maintenance, cleaning and disinfection, inspection of key components, usage standards, and long-term storage.
1、 Key points of daily maintenance
1. Appearance and Cleaning
-After each use, wipe the outer shell with a dust-free cloth to remove any spilled sample residue and avoid long-term adhesion of corrosive substances.
-Check for any damage to the power cord and plug to prevent the risk of electrical leakage; The plug needs to be cleaned regularly with alcohol swabs.
-To prevent liquid splashing on the control panel, dry cotton swabs can be used to clean the gaps and dust.
2. Lubrication of mechanical components
-Check the flexibility of the homogenizer head (or blade) weekly. If there is any jamming, apply food grade lubricating oil (such as silicone oil) to the rotating shaft.
-The pneumatic homogenizer needs to be checked for air leakage at the air circuit joints and regularly sealed with Vaseline to enhance air tightness.
3. Running status monitoring
-Run without load for 10 seconds, observe whether the motor makes any abnormal noise and whether the homogenization head shakes abnormally. If any abnormalities are found, immediately stop the machine and report for repair.
-Record the daily usage time, and after working for more than 8 hours, it is recommended to stop the machine and cool it down for 30 minutes to avoid overheating and damaging the motor.
2、 Deep cleaning and disinfection standards
1. Disassembly and cleaning of homogenization head
-Immediately remove the homogenizer head after use, soak it in a neutral detergent solution for 30 minutes, and gently brush the gaps with a soft bristled brush.
-Stubborn stains can be treated with an ultrasonic cleaning machine (frequency 40kHz, time 5 minutes), and strong acid/alkali solvents are prohibited.
2. Disinfection treatment
-Chemical disinfection: Wipe the surface of the homogenizer head with 75% ethanol and irradiate with ultraviolet light for 30 minutes; High temperature resistant components (such as stainless steel blades) can be sterilized under high pressure at 121 ℃ for 20 minutes.
-Biological pollution prevention and control: After processing pathogen samples, soak the homogenizer head in chlorine containing disinfectant (500mg/L) for 1 hour, rinse with water, and then dry.
3. Cleaning of chambers and pipelines
-Dismantle the equipment cover every month, use a vacuum cleaner to remove internal dust, and check if the ventilation openings are blocked.
-The gas pipeline of the pneumatic homogenizer needs to be backflushed with compressed air every quarter to prevent sample drying, clumping, and blockage.
3、 Special inspection of key components
1. Homogenization head and blade maintenance
-Check the wear of the blades every month, and replace them promptly if there are curled or chipped blades (it is recommended to backup blades of the same model).
-The tightness of the homogeneous head threaded connection should be checked after each use to prevent slipping caused by vibration loosening.
2. Motor and transmission system
-Remove the cover plate every six months, clean the dust on the motor carbon brush, and check the tightness of the belt (with a displacement of ≤ 2cm when pressing the middle of the belt with your fingers).
-High temperature resistant lubricating grease (such as Shell Alvania EP2) should be replenished annually in the bearing area to avoid dry wear and tear.
3. Sealing performance verification
-The pneumatic homogenizer needs to undergo regular pressure testing: adjust the pressure to 0.4 MPa, turn off the gas source, and hold the pressure for 10 minutes. If the pressure drops by more than 5%, the sealing ring needs to be replaced.
-Check the O-ring of the electric homogenizer for aging and deformation, and apply vacuum silicone grease to enhance the sealing performance.
4、 Operational norms and risk mitigation
1. Sample preprocessing requirements
-The sample needs to be centrifuged in advance to remove large particles of bone debris or hair, in order to avoid blocking the homogeneous gap.
-The liquid sample size should not exceed 2/3 of the capacity of the homogeneous cup to prevent overflow and corrosion of the motor.
2. Parameter setting restrictions
-Speed control: It is recommended that the handheld homogenizer should not exceed 15000rpm, and the desktop model should not exceed 80% of the nominal maximum speed.
-Cycle times: Low temperature sensitive samples should be homogenized continuously for no more than 30 seconds, with ice bath cooling intervals.
3. Emergency situation handling
-Stop running immediately when there is abnormal vibration and check if the sample is stuck in the homogenization head; Smoke or burnt smell indicates motor overload, power should be cut off and reported for repair.
-When liquid leaks onto the circuit board, immediately remove the power supply, wipe with anhydrous ethanol, and dry for 48 hours.
5、 Long term storage and restart preparation
1. Storage environment requirements
-When the equipment is not in use for more than one week, it needs to be cleaned and placed in a moisture-proof box with a built-in desiccant (humidity<40%).
-Apply anti rust oil to the homogenizer head and wrap the bearing area with cling film to prevent oxidation.
2. Check before restart
-After releasing the storage state, conduct a no-load test and measure the noise level (should be<60dB).
