【Anaerobic yolk agar baseUsage】

Used for the inspection and cultivation of Clostridium botulinum in imported and exported food (SN/T 0865-2000)

【 Principle of Inspection 】

Yeast powder, tryptone, and peptone provide nitrogen sources, vitamins, and growth factors; Glucose provides a carbon source; Sodium chloride is used to maintain cell osmotic pressure; Agar is a coagulant.

【Anaerobic yolk agar basic mediumFormula]

Formula (per liter) content

Yeast extract 5.0g

Tryptone 5.0g

Monthly peptone 20.0g

Sodium chloride 5.0g

Agar 15.0g

Distilled water 1000mL

Final pH 7.0 ± 0.2

【 Usage 】

1. Weigh 50 g of anaerobic yolk agar base medium, add 1000 mL of distilled water or deionized water, stir and heat to boil until dissolved, and sterilize at 121 ℃ for 15 minutes. Cool to around 50 ℃, add 10-15 mL of 50% sterile yolk saline suspension to every 100ml of culture medium, shake well, and immediately pour onto a plate.

2. Take an appropriate amount of fungal moss with an inoculation ring, gradually dilute it with sterile 0.85% physiological saline at a ratio of 1:100 or 1:100 to the appropriate concentration, inoculate by pouring, and incubate in a 35 ℃ -37 ℃ incubator for 24 hours.

3. Result observation attachment: 50% yolk solution: Take fresh eggs, wash the eggshells with a hard brush, drain them, soak them in a 70% alcohol solution for 1 hour, remove the yolk with sterile operation, add an equal amount of sterile physiological saline, mix well and set aside
