Analytical techniques for chromatographic sample bottles
Date: 2025-12-11Read: 2
The analysis techniques for chromatographic sample bottles cover multiple aspects such as material selection, pretreatment, filling operation, storage maintenance, and quality control troubleshooting. The following is a detailed analysis of these techniques:
1、 Material selection and certification standards
Glass material:
Class I borosilicate glass: resistant to temperatures exceeding 500 ℃, suitable for high-temperature headspace analysis.
Class II sodium calcium glass: requires silanization treatment to meet USP<660>surface inertness requirements.
Quartz glass: used for ultra trace element analysis, with iron content below 1ppm.
Polymer materials:
Certification mark: The bottom of the USP/EP certified bottle should be marked with 'USPTypeI' laser etched. Trace analysis bottles need to be tested for metal impurities by ICP-MS.
2、 Preprocessing standardization process
Third level cleaning procedure:
Initial cleaning: Use 1% Hellmanex III ultrasonic cleaning for 30 minutes (frequency 40kHz).
Precision washing: Use ultrapure water (18.2M Ω· cm) for gradient rinsing.
Passivation: Soak overnight in 5% nitric acid (ICP-MS special bottle).
Drying quality control:
Vacuum drying oven parameters: 60 ℃/12 hours/-0.1MPa.
Nitrogen purge flow rate: 5L/min, lasting for 10 minutes.
Moisture residue detection: Karl Fischer method detection, residual amount should be less than 50ppm.
Deactivation processing technology:
Gas phase silanization: using hexamethyldisilazane vapor deposition.
Plasma treatment: Treat with Ar/O ₂ mixed gas for 30 seconds.
3、 Filling operation specifications
Volume control principle:
Conventional analysis: The filling amount should not exceed 80% of the nominal volume.
Headspace analysis: The liquid loading error should be controlled within ± 1% (using Gilson liquid workstation).
Anti volatilization measures:
Volatile samples: Pre cooled to 4 ℃ and quickly filled using PTFE/silicone composite spacers (with no more than 3 punctures).
Supporting screw cap torque control: torque range of 0.9-1.2N · m (recommended to use TorqueMeter calibration).
Special sample processing:
Photo sensitive sample: Select amber bottles and wrap them with aluminum foil.
Oxygen sensitive sample: The liquid surface is covered with a high-purity nitrogen layer (purity ≥ 99.999%).
Suspension sample: pre mixed with 2mm glass microspheres.
4、 Storage and maintenance system
Environmental monitoring parameters: It is necessary to strictly monitor the storage environment parameters.
Life cycle management:
Turnover frequency: HPLC bottles should not exceed 20 times (TOC detection should be below 500ppb).
Mechanical strength testing: Regularly conduct water pressure testing (pressure ≥ 3bar, hold for 5 minutes).
Surface defect inspection: Use a 10x magnifying glass to observe scratches.
5、 Quality Control and Troubleshooting
Blank control test:
GC-MS full scan detection: scanning range m/z 50-650.
HPLC-ELSD baseline noise: should be below 20 μ V.
ICP-OES metal leaching amount: should be less than 10 ppb.
Common problem solutions:
Special management of headspace bottles:
Sealing test: using vacuum attenuation method (leakage rate should be less than 0.5mbar/minute).
Aluminum cover deformation degree: measured using a digital caliper (height change should be less than 0.1mm).
It is recommended to use a magnetic capping device in conjunction: the pressure uniformity CV should be less than 5%.
