What are the technical pathways for peptide lysing instruments?
Date: 2025-12-19Read: 0
Peptide lysis analyzerBy using physical, chemical, or biological methods to break peptide bonds in polypeptide chains, complex proteins are transformed into analyzable peptide segments.Peptide lysis analyzerIts technological path is mainly divided into three major systems:
enzymatic hydrolysis
Principle: By utilizing the specific cleavage ability of biological enzymes such as trypsin and pepsin, specific peptide bonds at the carboxyl end of amino acid residues (such as lysine and arginine) can be accurately identified and broken under mild conditions (such as 37 ℃ constant temperature).
Advantages: The fragment length of the lysate is uniform, suitable for peptide sequence analysis, mass spectrometry identification, and other scenarios. For example, trypsin can cleave bovine serum albumin into a peptide mixture with a theoretical coverage of over 80% within 16 hours.
Limitations: Sensitive to enzymatic hydrolysis conditions such as pH and temperature, and difficult to handle peptides containing large amounts of proline or modified amino acids.
Chemical cracking method
Principle: Molecular level cleavage is achieved under high temperature (100 ℃) conditions using strong acids (such as 6M hydrochloric acid) or strong bases (such as sodium hydroxide solution), suitable for difficult to degrade collagen, keratin, etc.
Advantages: High cracking efficiency, capable of handling complex samples.
Limitations: The reaction conditions are severe and may lead to non-specific cleavage or amino acid side chain modification, requiring subsequent purification steps.
Physical cracking method
Principle: Using lasers (such as 193nm excimer laser pulses) to generate local ultra-high temperatures within nanoseconds, causing non thermodynamic cleavage of peptide bonds.
Advantages: No chemical reagent residue, suitable for samples sensitive to chemical contamination (such as clinical diagnostic specimens).
Limitations: High equipment costs and strict technical requirements for operation.
