Experimental technology of fully automatic protein immunoblotting instrument
Date: 2025-08-22Read: 2
Fully automated Western blotting is an experimental technique widely used in biomedical, cell biology, and molecular biology research, primarily for detecting the presence, expression levels, and modification status of specific proteins. This technology combines electrophoresis separation and immunoassay, using specific antibodies to identify target proteins.
The basic working principle of the fully automatic protein immunoblotting instrument is as follows:
Sample preparation:
Total protein is extracted from cells, tissues, or body fluids, typically using lysis buffer for cell lysis.
The extracted protein is quantified using protein concentration determination methods such as BCA or Bradford assay.
SDS-PAGE electrophoresis:
The electrophoresis process separates proteins in the sample according to their molecular weight. Fully automated immunoblotting machines typically have automated electrophoresis capabilities, allowing for efficient and stable operation of SDS-PAGE electrophoresis.
After the electrophoresis process, the protein will migrate to the gel according to the size.
Film transfer:
Transfer the protein separated by SDS-PAGE onto a polyvinylidene fluoride (PVDF) membrane or nitrocellulose membrane. The membrane transfer process usually uses the electric transfer method to ensure the fixation of proteins on the membrane.
Sealing membrane:
The membrane after membrane transfer needs to be sealed with a blocking solution (usually 5% skim milk powder or BSA buffer) to block non-specific binding sites and prevent non-specific binding of antibodies to non target sites on the membrane.
Antibody incubation:
Incubate the membrane with primary antibodies (specific antibodies against the target protein). The incubation time and temperature need to be adjusted according to the requirements of the antibody.
Then, the membrane is incubated with a secondary antibody (usually an enzyme labeled antibody that binds to the primary antibody), which enhances the signal and makes subsequent detection more sensitive.
Signal detection:
Finally, the signal of antibody binding to the target protein is detected through substrate reactions (such as chemiluminescence or colorimetric methods), typically using chemiluminescence detection systems.
The automation system can automatically perform signal enhancement, exposure, and image capture according to the set time and intensity.
The characteristics and advantages of fully automatic protein immunoblotting instrument:
Efficiency and Automation:
The fully automatic protein immunoblotting instrument can complete a series of operations from sample loading, electrophoresis, membrane transfer, antibody incubation to detection results, greatly improving experimental efficiency and reducing errors caused by manual operations.
Precise control and stability:
The automation system can accurately control experimental parameters such as temperature, time, antibody concentration, etc., ensuring the repeatability and stability of each step.
Reduce human errors:
Automated instruments can reduce human intervention in experiments, ensure consistency in operations, especially in high-throughput experiments, and improve the reliability of results.
High throughput and multi sample processing:
The fully automatic immunoblotting instrument can process multiple samples simultaneously and is suitable for experiments that require high-throughput analysis, such as drug screening, clinical testing, etc.
Multi functional integration:
Many modern fully automated immunoblotting machines also integrate other functions such as electrophoresis, membrane transfer, antibody incubation, signal detection, etc. Users can customize operations according to their needs.
Application fields:
Protein expression level analysis:
Used for quantitative analysis of the expression levels of specific proteins in cells or tissues, especially in fields such as cancer and immune system research.
Protein modification research:
Detect post-translational modifications such as phosphorylation, acetylation, and glycosylation of proteins, and study how these modifications affect protein function.
Pathological research:
In disease diagnosis, Western Blotting can detect specific proteins associated with the disease, such as biomarkers for viral infections such as HIV and hepatitis B.
Drug development and screening:
In drug research, Western blotting is commonly used to evaluate the effects of drugs on the expression of specific proteins.
Basic scientific research:
Western blotting is a tool used in molecular biology research to study gene expression regulation, cell signaling pathways, and more.
Summary:
The fully automatic protein immunoblotting instrument automates traditional Western Blotting technology and has advantages such as high efficiency, precision, and reliability. It is widely used in life science research, medical diagnosis, and the pharmaceutical industry. Through automated operations, the throughput and repeatability of experiments have been greatly improved, human errors have been reduced, and a more stable experimental platform has been provided for researchers.
