In the field of plant cell culture, precise environmental control is the cornerstone of successful experiments. As core equipment, the constant temperature shaker and the light constant temperature shaker simulate and manipulate the in vivo environment of plant cells through precise control of temperature, oscillation, and light conditions, directly determining the fate of cell dedifferentiation, proliferation, and redifferentiation. This article will delve into the scientific principles behind these key configurations.

1、 Common configurations:

  • Temperature: 25 ℃

  • Rotation speed: 50-100rpm (low speed for fragile cells (50rpm), high speed for callus tissue blocks (100rpm))

  • Light intensity: 1000-3000 lux (most plants require dark cultivation to avoid light inhibiting dedifferentiation), differentiation stage (such as inducing bud/root formation): specific light conditions are required)

The key roles and configuration principles of constant temperature shaker and light constant temperature shaker in plant cell culture

  • Temperature control: why must it be 25 ℃

One of the core functions of a constant temperature shaker is to provide a constant and precise temperature environment. Almost all plant cell cultures set the temperature at 25 ℃, based on the following principle:

Optimal enzyme activity: 25 ℃ is the golden temperature for physiological and biochemical reactions in most temperate plants. At this temperature, enzymes and proteins involved in cell division, metabolism, and signal transduction have the highest activity and can effectively support rapid cell growth.

Avoid stress response: Excessive temperature (>30 ℃) can lead to protein denaturation; If the temperature is too low (<20 ℃), it will significantly reduce the metabolic rate. The constant temperature shaker eliminates temperature fluctuations, ensuring the repeatability and consistency of the experiment.

  • Speed selection: analysis of the difference between 50rpm and 100rpm

The rotational speed of the constant temperature shaker is not set arbitrarily, but is scientifically selected based on cell type and culture purpose.

Core function: The rotational speed provides two key functions - ventilation (ensuring oxygen supply and carbon dioxide excretion) and dispersion (preventing cell deposition and clumping, ensuring uniform distribution of nutrients).

50rpm (low speed) application: suitable for fragile cells (such as protoplasts). Low speed rotation can provide basic gas exchange while minimizing shear forces, protecting the integrity of cell membranes without cell walls, and preventing cell rupture.

Application at 100rpm (high speed): suitable for callus tissue blocks. High speed can generate stronger shear forces, effectively breaking down large cell clusters, increasing the contact surface for growth, preventing tissue settling, and thus achieving more efficient proliferation efficiency.

  • Lighting Strategy: The Irreplaceability of Light Controlled Temperature Shakers

Light is the core signal that regulates plant development, and this is precisely where the constant temperature shaker under light plays its irreplaceable role. It can combine the control of light intensity, light quality, and light period with the constant temperature oscillation function of the shaker.

1. Dark culture stage (dedifferentiation/proliferation)
At this stage, a regular constant temperature shaker is usually used for dark cultivation.

Principle: Darkness simulates a natural traumatic environment and is a strong signal that promotes cell dedifferentiation. At the same time, photosynthesis degrades key hormones (such as auxin 2,4-D) and inhibits cell division, promoting cell differentiation into chloroplasts, which goes against the goal of rapid proliferation.

2. Light culture stage (redifferentiation/organogenesis)
When it is necessary to induce rooting or sprouting, a light controlled shaker must be used to provide specific light conditions.

Principle: Light (especially blue and red light) is the core signal that triggers plant light morphogenesis. The constant temperature shaking table can accurately provide the required light intensity (1000-3000 lux) and light quality, activate light receptors (photosensitive pigments, cryptochrome), regulate endogenous hormone balance, and thus precisely command whether cells develop into roots or shoots.

  • How to choose between a constant temperature shaker and a light constant temperature shaker?

Choosing a constant temperature shaker: If your research mainly focuses on cell dedifferentiation, callus proliferation, or the establishment and maintenance of suspension systems, and the entire process requires dark culture, then a high-precision, stable shaking constant temperature shaker is your ideal choice.

Choosing a constant temperature shaking table under light: If your research involves stages that require light conditions such as plant organogenesis, rapid seedling propagation, photosynthesis research, or the production of secondary metabolites induced by light, then a constant temperature shaking table that can provide uniform and controllable light is suitableCore equipment.

By understanding these principles, you can better utilize them according to experimental needsConstant temperature shaker and illuminated constant temperature shakerTo optimize the plant cell culture process and obtain better experimental results.
