Introduction to the use of full temperature oscillation incubator
Date: 2025-08-29Read: 1
The full temperature oscillation incubator is a laboratory equipment that integrates temperature control, oscillation cultivation, and environmental simulation. It is widely used in experimental scenarios such as microbial cultivation, cell suspension cultivation, enzyme reactions, and strain screening. Its core advantage lies in the ability to accurately regulate temperature (usually within the range of -10 ℃~60 ℃ or wider) and coordinate oscillations at different speeds to provide a stable and dynamic cultivation environment for the sample. The following provides a detailed introduction to its usage from four dimensions: operation process, parameter settings, maintenance, and precautions.
1、 Preparation before operation: Safety and environmental verification
The preparation work before use is the foundation for ensuring the stable operation of the equipment and the safety of the experiment. The following three points should be focused on:
Equipment placement and power inspection
Place on a level, ventilated, and dry laboratory countertop, away from heat sources (such as alcohol lamps, ovens), water sources, and corrosive gases, and avoid direct sunlight. A space of ≥ 30cm should be reserved on both sides and the back of the device to ensure the normal operation of the cooling fan.
Confirm that the power supply voltage is consistent with the rated voltage of the equipment (usually 220V/50Hz), and that the power socket is reliably grounded to avoid leakage or unstable equipment operation.
Selection and loading of culture bottles/containers
Choose the appropriate container based on the oscillation mode (reciprocating/rotary): rotary oscillation is suitable for triangular flasks (50mL~2000mL), while reciprocating oscillation is suitable for culture dishes and rectangular culture bottles.
The sample loading capacity should be moderate: the liquid sample should not exceed 1/3~1/2 of the container volume (such as a 250mL triangular bottle with a maximum capacity of 100mL), to avoid liquid overflow and contamination of the equipment or instability of the center of gravity during oscillation.
If multiple containers are placed simultaneously, they should be evenly distributed on the oscillating tray, with symmetrical weight, to prevent the tray from tilting or generating severe vibrations during operation.
Equipment status check
Open the box door and check if the oscillation tray is stable and if the fixing screws are loose; Confirm that the surfaces of the temperature sensor and humidity sensor (if equipped with humidity function) inside the box are clean and unobstructed.
Check if the water level in the water tank (if it is a humidifying type) is between the "MIN-MAX" scale. If it is insufficient, add distilled water (avoid using tap water to prevent scale blockage).
2、 Core operation process: from startup to operation
The operation logic of the full temperature oscillation incubator follows the sequence of "parameter setting → sample placement → startup operation → process monitoring", with the specific steps as follows:
1. Familiarity with startup and interface
Press the "power switch" on the front of the device, the display screen will light up, and enter the main operation interface. The interface usually includes modules such as temperature setting, speed setting, time setting, and operating status (the layout of the interface may vary slightly among different brands, but the core functions are the same).
2. Place the sample and close the box door
Fix the container loaded with samples onto the oscillation tray using a fixture (the fixture needs to be tightened to prevent the container from sliding during oscillation);
Close the box door and confirm that the door seal is tightly attached (aging of the door seal can cause temperature leakage and affect control accuracy). Some models require pressing the "door control switch" to confirm that the door is closed properly.
3. Start up operation and process monitoring
Press the "Start" button on the interface, and the device will start running. The display screen will show real-time parameters such as current temperature, speed, and remaining time;
During the initial operation (first 30 minutes), close observation is required:
Whether the temperature has steadily increased to the target value (if the heating is too slow, check if the heating tube is working; if the cooling is too slow, check if the refrigeration system is lacking in fluorine);
Whether the oscillation is stable and whether there is any abnormal noise (if the noise is too loud, it may be due to tray imbalance or bearing wear);
Is there any overflow of the sample and is the container loose.
3、 After the experiment: shutdown and cleaning
Gradually shut down
First, adjust the oscillation speed to "0r/min", wait for the tray to completely stop, and then turn off the temperature control module;
If the refrigeration function is used, it is recommended to turn off the refrigeration first and let the equipment naturally return to room temperature (10-15 minutes) before cutting off the main power to avoid damage to the refrigeration system due to excessive temperature difference.
Sample and equipment cleaning
Take out the sample, clean the fixtures and residual liquid on the tray in a timely manner (if there is overflow, wipe it with a dry cloth, and stubborn stains can be disinfected with 75% alcohol);
Open the box door and allow the moisture inside to naturally dissipate, avoiding the growth of mold; Regularly clean the dust on the heat dissipation holes inside the box with a soft bristled brush to prevent poor heat dissipation.
follow-up inspection
Drain the remaining water in the water tank (when not in use for a long time) and clean the inner wall of the water tank;
Turn off the main power, unplug the plug, and cover the surface of the device with a dust-proof cloth.
4、 Safety precautions
Electricity safety: It is prohibited to plug in or unplug the power plug during equipment operation, and it is forbidden to operate the control panel with wet hands; Cut off the main power supply when not in use for a long time.
Load safety: Strictly comply with the rated load of the equipment (such as the maximum load capacity of 20kg on the pallet), prohibit overloading operation, and avoid motor burnout.
Operation standard: When opening the box door, reduce the speed or pause the oscillation to prevent sample splashing; When operating at low temperatures, avoid touching the refrigeration components inside the box directly with your hands (to prevent frostbite).
Emergency response: If there is any abnormal odor or smoke during operation, immediately press the "emergency stop" button (some models are equipped with it), cut off the power, contact professional personnel for maintenance, and prohibit self disassembly.
In short, the use of a full temperature oscillation incubator should follow the principles of "precise parameter setting, smooth start-up and operation, and standardized maintenance", and flexibly adjust key indicators such as temperature and speed according to experimental needs. At the same time, attention should be paid to daily inspection and safe operation of the equipment to ensure the reliability of experimental results and the service life of the equipment.
