Guidelines for the Application of Microbial Degradation Respirometers: Sample Preparation, Parameter Setting, and Data Analysis Techniques
Date: 2025-09-16Read: 4
Microbial degradation respiration apparatus is an important tool for studying microbial metabolic activity and degradation ability, widely used in fields such as environmental science, soil science, microbiology, and ecology. It evaluates the activity and degradation efficiency of microorganisms by measuring the amount of carbon dioxide released during the degradation of organic matter. This article will provide you with a detailed application guide for microbial degradation breathers, covering sample preparation, parameter setting, and data analysis techniques.
1、 Sample preparation: Ensure the reliability of experimental results
(1) Collection and processing of soil samples
Soil is the most common sample type in microbial degradation research. When collecting soil samples, representative locations should be selected to avoid areas that are contaminated or disturbed. The collected soil samples should be processed as soon as possible to prevent changes in microbial activity. Usually, soil samples need to be air dried or freeze-dried to remove moisture and maintain microbial activity. During air drying, direct sunlight should be avoided to prevent excessive temperature from affecting the activity of microorganisms. Freeze drying can better maintain the activity of microorganisms in the soil, but at a higher cost.
(2) Addition of organic substances
In order to study the degradation ability of microorganisms towards specific organic substances, it is usually necessary to add organic substances to soil samples. These organic substances can be natural, such as plant residues, animal feces, etc., or artificially synthesized, such as plastics, pesticides, etc. When adding organic matter, it should be ensured that it is evenly distributed in the soil sample. Uniform distribution can be achieved through stirring or mixing. The amount added should be adjusted according to the experimental purpose and soil properties, usually expressed as a percentage of dry weight.
(3) Sample pretreatment
Before conducting microbial degradation experiments, samples usually need to be pre treated. The purpose of preprocessing is to remove impurities that may interfere with experimental results, such as inorganic carbon, heavy metals, etc. For example, carbonate in soil can be removed by acid or alkali washing to avoid interference with carbon dioxide measurement. After pretreatment, it should be ensured that the pH value and ionic strength of the sample are restored to an appropriate range to ensure the normal growth and metabolism of microorganisms.

2、 Parameter setting: Optimize experimental conditions
(1) Temperature control
Temperature is an important factor affecting microbial metabolism and degradation activities. Most microbial degradation breathers are equipped with temperature control systems that can set and maintain a constant temperature. When setting the temperature, it should be adjusted according to the optimal growth temperature of microorganisms and the experimental purpose. For example, for soil microorganisms, it is usually set between 25 ℃ and 30 ℃. For some microorganisms in the Ji environment, higher temperatures may need to be set. The stability of temperature is crucial for the reliability of experimental results, so the temperature control system should be regularly checked to ensure its normal operation.
(2) Humidity control
Humidity also affects the metabolic activity of microorganisms. Excessively dry or humid environments can inhibit the growth and degradation ability of microorganisms. In the experiment, the moisture of the soil sample should be adjusted by adding an appropriate amount of water. Usually, the moisture content of soil samples should be maintained between 60% and 80% of the field capacity. Humidity can be monitored and regulated through weighing methods or soil moisture sensors. Regularly add moisture to maintain a suitable humidity level and avoid humidity drops caused by water evaporation.
(3) Gas exchange
Microorganisms consume oxygen and release carbon dioxide during the degradation of organic matter. Therefore, gas exchange within the reaction vessel is crucial for maintaining the normal metabolism of microorganisms. In the experiment, it should be ensured that the gas inside the reaction vessel can exchange freely. Gas exchange can be achieved by installing gas vents on the container or using breathable sealing materials. At the same time, external gas pollution such as carbon dioxide, hydrogen sulfide, etc. should be avoided, as these gases may interfere with the experimental results. Regularly replacing gases or using gas purification devices can ensure the quality of gases.
3、 Data analysis: extracting valuable information
(1) Calculation of respiratory rate
The microbial degradation respiration meter evaluates the metabolic activity of microorganisms by measuring the release rate of carbon dioxide. The respiratory rate is usually expressed as the amount of carbon dioxide released per unit time and per unit mass. When analyzing data, the respiratory rate should be calculated based on the readings of the instrument and the weight of the sample. For example, if the instrument displays a release of 10 microliters of carbon dioxide per minute and the sample weight is 10 grams, the respiration rate is 1 microliter of carbon dioxide per minute per gram. The change in respiratory rate can reflect the strength of microbial metabolic activity and the progress of degradation processes.
(2) Assessment of degradation efficiency
By analyzing the changes in respiratory rate over time, the degradation efficiency of microorganisms towards organic matter can be evaluated. Usually, the respiratory rate increases rapidly in the early stages of the experiment, and gradually decreases as organic matter degrades. By plotting the curve of respiratory rate over time, the starting, peak, and ending points of the degradation process can be determined. The degradation efficiency can be evaluated by calculating the difference between the initial amount and the final residual amount of organic matter. For example, if 100 milligrams of organic matter are initially added and 20 milligrams remain after the experiment, the degradation efficiency is 80%.
(3) Statistical analysis of data
To ensure the reliability of experimental results, statistical analysis should be conducted. Methods such as one-way analysis of variance (ANOVA) or t-test can be used to compare the differences between different treatment groups. Statistical analysis can help determine the significance of experimental results and avoid misjudgments caused by random errors. At the same time, statistical indicators such as mean, standard deviation, and coefficient of variation should be calculated to evaluate the dispersion and reliability of the data. Through statistical analysis, it is possible to more accurately evaluate the degradation ability of microorganisms and optimize experimental conditions.
4、 Summary
Microbial degradation respiration apparatus is a powerful tool used to study the metabolic activity and degradation ability of microorganisms. By using correct sample preparation, parameter setting, and data analysis techniques, the reliability and accuracy of experimental results can be ensured. In sample preparation, the representativeness of soil samples and the uniform distribution of organic matter should be ensured. In parameter settings, temperature, humidity, and gas exchange conditions should be optimized to maintain the normal metabolism of microorganisms. In data analysis, respiratory rate and degradation efficiency should be accurately calculated, and statistical analysis should be conducted to evaluate the reliability of the data. Through these techniques, researchers can better utilize microbial degradation respirators to provide scientific basis for environmental protection and sustainable development.
