Taking the LGJ-12A freeze dryer from Beijing Fourth Ring Road as an example, during pre freezing, the sample can be placed in a cold trap at room temperature and pre frozen for 4 hours to solidify.

If the sample is frozen in a -80 ℃ low-temperature refrigerator, the temperature of the cold trap needs to be lowered to -50 ℃ first, and then the sample can be placed directly and vacuum dried.

If the sample is frozen in a -20 ℃ low-temperature refrigerator, the temperature of the cold trap should be lowered to below -20 ℃ first, and then the material should be taken out of the refrigerator and frozen for another hour before drying can begin.

For the pre freezing stage, if the container is a centrifuge tube, a 50ml centrifuge tube filled with 20-25ml of sample is sufficient. When freezing and pre freezing, the lid should be tightened, and the centrifuge tube should be placed horizontally for freezing. After four hours of pre freezing, drying can begin. If the container is a culture dish, it can be filled with an appropriate amount of sample, but be careful not to fill it too full, and there is no need to seal it during pre freezing.

Entering the drying stage, the centrifuge tube container can unscrew the bottle cap and place the centrifuge tube vertically into the freeze dryer.
