Peptide lysimeter is an instrument used in fields such as biochemistry, drug development, proteomics, etc. It is mainly used to break polypeptide chains into smaller fragments for subsequent analysis.
The hardware composition of a peptide lysis instrument usually includes the following core parts:
1. Sample processing system
Sample loading module: used to load peptide samples, usually including sample tubes, syringes, or automatic samplers.
Reaction chamber: The sample undergoes cracking reaction in the reaction chamber, usually made of corrosion-resistant materials (such as stainless steel or inert alloys), and has temperature control function.
Stirring or mixing device: used to ensure that the sample is thoroughly mixed with the lysis reagent, which may include a magnetic stirrer or ultrasonic mixing device.
2. Cracking reaction system
Cracking reagent supply module: used to provide the reagents required for cracking reactions (such as enzymes, strong acids, strong bases, or other chemical reagents), usually including reagent storage tanks and precise pumping systems.
Reaction control module: used to control the conditions of cracking reaction (such as temperature, time, pH value, etc.), usually equipped with heating or cooling devices (such as Peltier temperature control system).
Reaction termination module: used to terminate the reaction after the cracking reaction is completed, which may include neutralizing reagents or rapid cooling devices.
3. Separation and Purification System
Chromatography column or separation device: used for the separation and purification of peptide fragments after fragmentation, which may include high-performance liquid chromatography (HPLC) columns or capillary electrophoresis devices.
Detector: Used to monitor separated peptide fragments, such as ultraviolet detectors (UV), fluorescence detectors, or mass spectrometry interfaces.
4. Peptide lysis analyzer control system
Temperature control module: used to accurately control the temperature of the reaction chamber, usually using PID temperature control technology.
Pressure control module: used to control the pressure inside the reaction chamber to ensure that the reaction proceeds within a safe range.
Time control module: used to set the time for cracking reaction, usually implemented by a programmable logic controller (PLC) or software.
5. Data Collection and Analysis System
Data acquisition module: used to record parameters during the cracking reaction process (such as temperature, pressure, time, etc.) and the detection results after separation.
Software system: Used for controlling instrument operation, data analysis, and result output, typically equipped with user-friendly interfaces and data processing tools.
6. Auxiliary system
Cleaning and maintenance module: used for cleaning reaction chambers and pipelines to prevent cross contamination, usually including automatic cleaning programs and waste liquid treatment devices.
Safety protection devices: such as leak prevention design, emergency stop button, protective cover, etc., to ensure safe operation.
Power and Communication Module: Used for power supply and data transmission, may include Ethernet, USB, or RS-232 interfaces.
7. Other optional components
Automation modules, such as automatic samplers, robotic arms, etc., are used to achieve high-throughput sample processing.
Mass spectrometry interface: used to directly import the cleaved peptide fragments into a mass spectrometer for further analysis.
Freeze drying device: used to process cracked samples for long-term storage or subsequent analysis.
