1、 Optimize core objectives
Multi channel peptide synthesizerThe core advantage lies in "parallel synthesis and batch production", and efficiency optimization needs to focus on "improving the success rate of single channel synthesis, shortening the synchronization period of multiple channels, and reducing side reaction losses". While ensuring peptide purity (usually ≥ 95%), it can achieve more high-quality peptide synthesis output per unit time while controlling reagent consumption costs.
2、 Optimization of synthesis parameters: precise regulation to improve reaction efficiency
1. Optimization of amino acid activation parameters
Activation reagent ratio and concentration: Adjust the activation system according to the type of amino acid (hydrophobic/hydrophilic, alpha helix/beta folding tendency). For example, when synthesizing insoluble amino acids (such as valine and leucine), use the HBTU/HOBt/DIEA activation system. The molar ratio of activation reagent to amino acid is increased to 1.2:1.0, and the concentration is controlled at 0.2-0.4mol/L to ensure sufficient activation; Conventional amino acids can be obtained using the DIC/HOBt system, with a molar ratio of 1.0:1.0 to meet the requirements and reduce reagent waste.
Activation time control: The conventional activation time for amino acids at room temperature is 3-5 minutes. Amino acids containing β - branches or aromatic side chains (such as isoleucine and phenylalanine) need to be extended to 5-8 minutes to avoid incomplete activation leading to a decrease in condensation efficiency; After activation, the feeding should be completed within 30 minutes to prevent the degradation of activated intermediates.
2. Optimization of condensation reaction parameters
Reaction temperature and time: Conventional peptide synthesis uses room temperature (25 ± 2 ℃) reaction, with a single step condensation time of 20-30 minutes; For peptides with steric hindrance (such as sequences containing multiple proline and tert butyl protecting groups), the temperature can be raised to 37 ℃ and the condensation time can be extended to 40-60 minutes, or a segmented heating mode (room temperature for 10 minutes first, then 37 ℃ for 30 minutes) can be used to improve the reaction conversion rate.
Solvent selection and dosage: When synthesizing hydrophilic peptides using DMF as the base solvent, 10% -20% DMSO can be added to enhance solubility; When synthesizing multiple channels synchronously, the amount of solvent used in each channel needs to be precisely controlled at 5-8 times the volume of the resin to ensure sufficient swelling of the resin, uniform contact between the reagent and the resin, and avoid incomplete local reactions due to insufficient solvent.
3. Optimization of deprotection parameters
Concentration and time of deprotection reagent: 20% piperidine/DMF solution is used as the deprotection reagent, and the conventional sequence deprotection time is 10-15 minutes; For amino acids containing difficult to deprotect groups such as Fmoc Arg (Pbf) and Fmoc Asn (Trt), the concentration of piperidine can be increased to 25%, or deprotection can be carried out in two stages (the first 5 minutes, followed by adding new reagent after discarding the solution and deprotection for another 10 minutes), to ensure a deprotection rate of ≥ 99% and avoid residual protective groups affecting subsequent condensation.
Optimization of washing process: After deprotection, wash 3-5 times with DMF, with a washing volume of 6 times the resin volume each time, and a washing time of 30 seconds per time. At the same time, enable multi-channel synchronous stirring function to improve washing efficiency and reduce amino acid racemization caused by pyridine residue.
3、 Multi channel collaborative optimization: avoiding synchronization bottlenecks
1. Channel load balancing design
According to the length and difficulty of peptide sequences, channels are allocated by crossing short sequences (≤ 10 amino acids) with long sequences (≥ 20 amino acids), easy to synthesize sequences, and difficult to synthesize sequences to different channels, in order to avoid a single channel slowing down the overall synthesis cycle due to long reaction times. For example, in 4-channel synthesis, 2 channels synthesize 15 peptides (conventional difficulty), 1 channel synthesizes 25 peptides (high difficulty), and 1 channel synthesizes 8 peptides (easy to synthesize), achieving approximate synchronization of multiple channels through reasonable allocation.
2. Optimization of reagent supply and distribution
Pre mixed reagent preparation: When synthesizing peptides of the same type in bulk, pre mix the activation reagent and amino acid solution according to the number of channels. Use specialized reagent bottles to separate and connect them to each channel to avoid wasting time on single channel preparation and ensure consistent concentration of reagents in each channel.
Pipeline and valve inspection: Before synthesis, check whether the multi-channel reagent distribution pipeline is unobstructed, whether the electromagnetic valve switch response is sensitive, and ensure that the feeding error of each channel reagent is ≤± 2%; For easily crystallizable reagents (such as HBTU), an insulation sleeve (temperature 25-30 ℃) should be wrapped around the reagent bottle to prevent crystallization from blocking the pipeline and causing uneven feeding.
3. Gradient heating and synchronous control
For the case of delayed reactions in some channels in multi-channel synthesis, a gradient heating strategy can be adopted: the temperature of the slower reaction channels is separately raised (not exceeding 40 ℃), and the "priority feeding of delayed channels" mode is set through the operating software to save 1-2 minutes for delayed channels in the early deprotection and washing steps, ensuring that the final multi-channel completes the condensation reaction synchronously and reducing waiting time.
4、 Optimization of equipment and reagent management: laying a solid foundation for efficiency
1. Equipment pre-processing and maintenance
Before synthesis, enable the preheating function of the equipment to stabilize the temperature of the reaction chamber at the set value (± 0.5 ℃), and calibrate the multi-channel stirring blades to ensure that the stirring speed of each channel is consistent (error ≤ 5rpm), avoiding local reaction efficiency differences caused by uneven stirring.
Regularly (every 50 batches of synthesis) clean the reagent distribution pipeline and reaction kettle, wash them alternately with DMF and methanol three times, remove residual amino acids and reagent precipitates, prevent pipeline blockage or cross contamination, and ensure the efficiency of reagent transportation.
2. Reagent quality and storage management
Select high-purity reagents (amino acid purity ≥ 99%, activation reagent purity ≥ 99%) to avoid impurities causing increased side reactions; Reagent storage should be classified according to requirements: solid amino acids should be sealed and frozen at -20 ℃, liquid reagents (such as piperidine, DMF) should be stored at room temperature in the dark, and used up within 3 months after opening.
Before use, check the status of the reagent: the water content of DMF should be ≤ 0.05% (which can be detected by Karl Fischer method), and the activation reagent should have no clumping or discoloration. If the reagent is not qualified, it should be replaced in a timely manner to avoid a decrease in synthesis efficiency or failure due to reagent problems.
5、 Special strategy for optimizing the efficiency of difficult to synthesize sequences
Hydrophobic peptides: A "reverse phase solvent system" (DMF/dichloromethane=7:3) is used to enhance the solubility of peptides, while adding 0.1% trifluoroethanol as a co solvent; The "dual activation" method is used during condensation (HBTU activation first, followed by adding a small amount of PyBOP to enhance reaction activity), and the single step condensation time is extended to 60 minutes.
Containing multiple repeated amino acid sequences, such as poly Gly and poly Ala, an additional "pre washing" step (washing the resin twice with DCM) is added before feeding the repeated amino acids to reduce the residual protective group fragments on the resin surface; Simultaneously reduce the condensation temperature to 20 ℃ to avoid amino acid racemization caused by high temperature.
Cyclic peptide synthesis: In the cyclization step, a "high concentration reaction system" (resin concentration increased to 0.1g/mL) is used, and a catalytic amount of HOAt (0.1eq) is added. The cyclization reagent is slowly added dropwise for 10-15 minutes to improve the cyclization efficiency and reduce linear peptide residue.
6、 Optimization effect verification and adjustment
After each optimization, the effectiveness needs to be verified through three indicators: ① synthesis efficiency (the number of qualified peptide batches completed per unit time); ② Peptide purity (detected by HPLC, target purity ≥ 95%); ③ Reagent consumption per 1mmol of peptide synthesis. If a certain indicator is not met, targeted parameter adjustments are needed: if the purity is insufficient, optimize the deprotection or condensation time; if the efficiency is not improved, check the channel load distribution or equipment operation status to ensure the effective implementation of the optimization plan.
