The following are detailed instructions for using a refrigerated Raman spectrometer:
1、 Preparation before use
1. Environmental control: It is necessary to avoid operating instruments in humid or high-temperature environments, as well as various magnetic field interferences, to ensure detection accuracy. The instrument should be placed in a dry, stable, and windless environment, with an ideal operating environment of temperature 15-30 ℃ and humidity ≤ 60% to prevent damage to the optical components.
2. Instrument status check: Before starting up, check whether all components are intact, including laser light source, spectrometer, analysis software, etc. After connecting the power supply, observe whether the indicator light is on normally, and confirm that the instrument has been preheated to the appropriate temperature and stabilized.
3. Sample pretreatment: The surface of the sample should be smooth and free of pollutants, avoiding background noise caused by oil stains, dust, or fingerprint residue. Solid samples can be directly placed on the sample stage, liquid samples require a dedicated liquid cell or capillary tube, and gas samples need to be introduced into the sample chamber through a specific interface. If necessary, it can be cleaned with solvent and dried.
2、 Core operational steps
1. Optical path adjustment and calibration: Adjust the laser beam to be correctly incident on the sample, and adjust the optical path for collecting scattered light to pass through the spectral system of the spectrometer. Use standard samples for wavenumber calibration to ensure the accuracy of the obtained spectral data; Use white samples for intensity calibration to ensure accurate comparison of light signals between different samples.
2. Parameter setting: Select the appropriate laser wavelength and power based on the characteristics of the sample, and set the corresponding spectral wavelength range, step size, and integration time parameters. Generally speaking, a lower laser power should be used for preliminary testing to avoid sample burning.
3. Data collection: Start the instrument for measurement and record the spectral signal reflected back from the sample. During the collection process, it is necessary to maintain a stable environment and avoid vibration and noise interference.
3、 Data Processing and Analysis
1. Background subtraction: Before testing, blank background signals need to be collected and subtracted during data processing to eliminate dark current and stray light interference.
2. Spectral analysis: Combining standard spectral libraries or literature data for peak analysis and spectral plotting, determine the position and intensity of Raman peaks, and avoid misjudging peak assignment.
4、 Daily maintenance and safety matters
1. Equipment maintenance: Regularly clean optical components, wipe lenses and filters with dust-free paper and isopropanol to avoid scratches or contamination; Avoid long-term high-power operation of the laser and regularly check the attenuation of the laser output power; Timely upgrade the instrument control software.
2. Safety protection: Follow laboratory safety regulations, wear goggles and gloves; Avoid looking directly at the laser path to prevent eye damage.
3. Long term storage: If not used for a long time, the main power switch should be turned off, the head socket should be unplugged and wrapped in packaging to prevent dust from entering. Neutral cleaning agents should also be used to clean the air conditioning filter.
Strictly following the above steps and precautions when operating a refrigerated Raman spectrometer can not only obtain high-quality spectral data, but also extend the service life of the equipment.
