Precautions for planning and decoration of sterile laboratory
Date: 2025-06-25Read: 1
sterile laboratoryThe planning and decoration of the laboratory must strictly follow core principles such as cleanliness, airflow control, material selection, and optimized operating procedures to ensure that the experimental environment meets the high-precision experimental requirements of microbiology, molecular biology, and other fields. The following are the key considerations from the three stages of design, construction, and acceptance:
1、 Design phase: Functional zoning and cleanliness planning
Classification of Cleanliness Levels
Determine the cleanliness level based on experimental requirements (such as ISO level 5/100, ISO level 7/10000), and clarify the difference in cleanliness between core operating areas (such as cell culture rooms, PCR laboratories) and auxiliary areas (such as buffer rooms, changing rooms).
Example: The PCR laboratory needs to reach ISO level 5, while the sample receiving area can be relaxed to ISO level 7.
Airflow organization design
Adopt unidirectional flow (laminar flow) or non unidirectional flow (turbulent flow) systems, with priority given to vertical unidirectional flow (wind speed 0.3-0.5m/s) in the core area to avoid cross contamination.
Key point: The return air outlet should be located away from the control panel to prevent airflow short circuits; The pressure gradient should ensure that the pressure in the core area is higher than that in adjacent areas (such as a pressure difference of ≥ 5Pa between the buffer zone and the core area).
Functional zoning and route planning
The principle of three zones and two buffers: strictly separate the clean zone, semi polluted zone, and polluted zone, and set up buffer rooms and airlocks.
Example: Personnel move from the cleaning area (changing room) → buffer room → core area, and materials are transferred through a transfer window (with UV sterilization function).
2、 Construction phase: Material and process control
Selection of enclosure structure materials
Wall and ceiling: Made of color steel plate (rock wool sandwich) or electrolytic steel plate, with a smooth surface, corrosion resistance, and easy cleaning.
Ground: Use PVC roll or epoxy self leveling, seamless splicing, anti slip and wear-resistant.
Avoid using ordinary latex paint or ceramic tiles, as they are prone to dust accumulation and difficult to disinfect.
Sealing and leak proof treatment
All gaps (such as color steel plate joints, doors and windows) need to be sealed with sealant or silicone strips to ensure airtightness.
Testing standards: After completion, a pressure difference test (pressure difference between the core area and adjacent areas ≥ 5Pa) and an air tightness test (air leakage rate ≤ 2%) are required.
Water, electricity and ventilation system
Circuit: Adopting concealed installation method, sockets and switches are designed with dust-proof and waterproof features to avoid exposed wires.
Water supply and drainage: The pipeline needs to be treated with anti-corrosion measures, and the drainage outlet should be equipped with a water trap and anti backflow device.
Ventilation system: HEPA filters need to be replaced regularly (recommended every 1-2 years), and duct connections should be reinforced with sealing tape.
3、 Acceptance and maintenance phase: performance validation and long-term management
Cleanliness verification
Use a laser particle counter to detect suspended particle concentration (ISO Level 5 requires ≥ 0.5 μ m particles ≤ 3520 particles/m ³).
Key point: During acceptance, it is necessary to simulate actual working conditions (such as personnel and equipment operation) to ensure that the dynamic cleanliness meets the standard.
Microbial monitoring
Regularly detect settling bacteria or planktonic bacteria (ISO level 5 requires settling bacteria to be ≤ 1CFU/dish · 4h), and establish microbial monitoring records.
Example: Surface wiping samples are taken weekly in the core area, and air samples are taken monthly.
Daily maintenance standards
Cleaning and disinfection: Use 75% ethanol or hydrogen peroxide disinfectant, and avoid using chlorine containing disinfectants (which can corrode equipment).
Equipment management: Regularly calibrate temperature and humidity meters, differential pressure meters, and replace aging components (such as seals and filters).
Personnel training: Develop SOP (Standard Operating Procedure) to standardize the procedures for changing clothes, disinfection, and experimental operations.
4、 Common Problems and Solutions
problem reason Solution
The cleanliness does not meet the standard Filter blockage, airflow short circuit Replace the HEPA filter and optimize the position of the return air outlet
pressure difference anomaly Insufficient airtightness, fan malfunction Re seal the joint and inspect the fan
excessive microorganisms Improper personnel operation and incomplete disinfection Strengthen personnel training and increase disinfection frequency
Ground warping or cracking Poor material quality and construction technology issues Select anti-static PVC rolls and standardize the construction process
Modular design: using detachable color steel plates for easy later modification. 5、 Suggestions for balancing cost and efficiency
Energy saving measures: Use variable frequency fans and LED lighting to reduce energy consumption.
Intelligent management: Integrated real-time monitoring system for temperature, humidity, pressure difference, and particle concentration to improve operation and maintenance efficiency.

summary
sterile laboratoryThe planning and decoration of the project should focus on cleanliness, airflow control, and material durability as the core, combined with optimized design through experimental processes, and ensure long-term stability through strict acceptance and maintenance. It is recommended to entrust a professional organization for design and construction, and refer to national standards such as the "Code for Design of Cleanrooms" (GB 50073).
